Flavour and Fragrance

QZyme WorkbenchTM has a unique functionality- QZyme PilotTM and QZyme ModellerTM to discover new enzymes. This computer driven enzyme discovery protocol finds suitable enzymes for a particular reaction without conflicting any IP issues.

Problem statement/Challenge

A chemical reaction was provided by the customer where a biotransformation was required. Customer was looking for a solution to reduce the side products that are generated during the chemical transformation. The reaction scheme was very unique as all the reactants and products are organic and not soluble in water medium. The task for Quantumzyme was to identify a suitable enzyme that has the substrate selectivity as well as it can catalyze the chemical reaction in organic medium.

Solution

Enzyme discovery platforms QZyme PilotTM and QZyme ModellerTM were used to identify the right enzyme. Lab validation was done to check if the product is forming or not after identifying the right enzyme. It turns out that the selected enzyme is able to catalyze the reaction without giving any side products. The product was identified by GC analysis. The entire journey starting from enzyme discovery to lab validation showed Quantumzyme’s capability to identify the suitable enzyme for a completely unknown reaction in the area of biocatalysis.

Problem statement/Challenge

Customer had shared a reaction which is catalyzed by a fungal species via the fermentation process. For less than 5 gm/L substrate loading, conversion is good. However, the conversion decreases with increasing substrate loading more than 5 gm/L.

Solution

Quantumzyme, as a solution provider, first identified the root cause of the problem. The challenge is the toxicity of the substrate towards the fungal species as well as the solubility of the substrate. To overcome the challenge related to toxicity, Quantumzyme has identified the responsible enzyme among more than 10,000 enzymes present in that strain and developed a cell free assay.

Enzyme Development for API synthesis

Most of the APIs are chiral and only one enantiomer is pharmaceutically active. The synthesis of enantiopure chiral molecules via chemical method is challenging. Quantumzyme has identified enzymes and engineered the identified enzyme for the synthesis of such APIs.

Textile Industry

Application of enzymes in denim washing for appearance and softening has been a common practice recently. Cellulases are widely used enzymes for denim washing but they are acid based and cause back staining. Alkaline stable Endoglucanases are a very good alternative for cellulases, however they lack adequate enzyme activity leading to high enzyme loading. At Quantumzyme, we have engineered such endoglucanases by studying the catalytic mechanism and binding of polymeric substrates with Endoglucanases. QZyme Hotspots and QZyme Designer helped in finding the mutable amino acid positions and effect of the change in amino acids. Thus, Quantumzyme improved the endoglucanase activity enabling commercial viable process of denim washing.
